I've been growing Oyster, Shiitake, and Lion's Mane mushrooms for around 6-7 years now and also grow a lot of cacti from seed. I've been meaning to get into TC for some time, considering I already have most of the gear, materials, and experience with very similar culture work and it's essentially a combination of two of my existing hobbies. Now I just need to clear out some space for cultures. Thanks for the video, it's a good intro into TC and helped solidify my understanding of it.

Dude you rock, All the other Tissue culture videos feel like armature vlog videos, or they explain well enough what Tissue culture is, but give no instructions for doing it. Your videos are put together so well and they convey the information perfectly! Thank you for making this information so easy to follow and understand! I've done fairy castle cactus and some succulents (echeverias and graptopetalums). Cacti grow well in TC

@@plantsinjars I’m mostly interested in extremely xerophytic cacti with tap roots. Something like Ariocarpus, Lophophora, Astrophytum caput-medusae, Pterocactus, etc. They are highly sensitive to moisture and are known to absorb water until they split open, which leads to disfigured specimens and rot. They also struggle to regrow a tap root from cuttings, which affects theirs looks, value, and ability to withstand drought. I’m curious how they cope with tissue culture considering the constant high humidity in the box, and whether they can grow a true tap root. Might have to give it a try myself. You’re definitely inspiring me to experiment.

@@morningstar8187lophophora and ariocarpus have been done TC. Also they’re more hardy than most give them credit. I grew lophophora grafting on periskiopsis for years hydroponicly and didn’t have issues with splitting. A friend of mine is a grower for NAC and does ebb and flow setup for thousands of lophophora

The bluelab pen I have now has worked well for the past year. I broke a bunch of other ones before that. Unfortunately IMO it’s super overpriced

I use the “Apera” brand, they make a replacement probe pen as well. It’s about 1/2 the price of bluelab, and i personally think equal in quality. I’m also using it daily, and many times daily. I have had the same pen for about 2 years and never had an issue. I will say that getting the PH pen storage solution, and weekly, if not daily calibration is a great way to extend the life.

If I am using a plant already in tc, and put it in multiplication media, do i need to cut it up, or put the whole plant in? First timer so figured i'd take your advice and start with a plant already in tc.

So, I’ve been using ketchup cups with mushroom cultures for a few years now. Does this still apply for this side of the fence? Just wondering about the size. I have glass, but I usually start in the cups and make the needed transfers until bobs my uncle.

If they’re autoclavable, they’ll work. Although you might find ketchup cups are a little too short for some plants in tc

This is alot to grow such easy to propagate plants like monstera I use Coco coir and perlite with a mix of nutrients and occasionally ga3 to propagate almost every plant your cuttings are big enough to go straight into a organic medium they will do much better aswell you won't be needing to wait so long for roots to grow just keep them in a container with high humidity I have my lights on 18/6 and I keep my fluorescent lights much closer to my cuttings then you I don't get contamination at all only time I use agar is for meristem culture which I would clean the plant material with a good blast of tap water then soak in a bit of hydrogen peroxide and water for 30mins then I strip most of the plant material away and just do meristem culture but that's for saving diseased plants but not just propagating them your set up is actually much better for mycology purposes you would do really well growing mushrooms

With your method, 1 node = 1 plant. With tissue culture, you can get 10+ plants per node. It’s more efficient

I don't think you're the target audience for TCs 🫠 they're used to mass produce plants. Mainly for nurseries. Imagine how painfully slow we would get plants on market if we had to wait for them to grow enough nodes to prop then grow and prop and grow and prop. Nothing would ever make it on the market. TCs are far more efficient and it's thanks to them we have the cooler variegated stuff.

I used to be a forensic scientist who attended clandestine drug laboratories. One of the more interesting ones was where a guy was growing magic mushrooms from spores in much the same way as you outlined. I think it was a penesis species but i was only qualified to confirm the chemical component psilocin. They were obviously mushrooms but i had to write something like "the vegetable matter contained psilocin" from memory. We were only able to report what we could attest was fact.

You definitely described it very simply and in detail. But i just don't understand why you wouldn't just grow the plant normally. I would really like to understand the benefits of TC versus growing a plant from seed or cutting

You can grow a much higher quantity of plants with tc than traditional methods of asexual cloning like taking cuttings
