Protoplast culture method | Easy explaination | Bio science
Protoplasts are cultured either in semisolid agar or liquid medium. Sometimes, protoplasts are first allowed to develop cell wall in a liquid medium and then transferred to agar medium. Agarose is the most frequently used agar to solidify the culture media.

Yes it does but many plants are polyploid, so it really isn't a problem. The correct term for this sort of polyploidy would be allopolyploidy.
