Fantastic vid, thanks. One small note about size-exclusion chromatography: The larger fragments travel FASTER. The smaller fragments travel SLOWER, as they are trapped in the porous beads in the column.

I have a suggestion, I see a pop up on your videos saying "Do you want me to do your homework for you?". I think you have made a good reputation, but by offering to "do" other students work is not ethical. I think you should keep on teaching students how to "catch" fish instead of "giving" fish if you really want to make a change. Thank you. Keep up the good work!

What happens if a tRNA, rRNA or even a mRNA has a sequence of consecutive A nt's in the middle of the chain. Will it end up attaching to the bead T-chain adaptors? Or does this method require a RNA chain to have A's only at the end?

Great video! But, can someone explain to me why Shomu lists SNP/mutation detection as a pro for RNA sequencing? I don't see why you couldn't attain the same result, perhaps even more accurately, by sequencing DNA in that case.

RNA-seq limits you to only sequencing the genes, so it can be cheaper than whole genome sequencing if you are only interested in gene sequences. Although exome sequencing can also be a good option if you only want SNPs within the genes and aren't interested in the expression levels at all. Also if you are already doing differential expression analysis, the SNPs are also a nice bonus analysis that you can do with the same RNA-seq data.
