If you are referring to Ouma calling Saihara “Saihara-chan”, that is canon. In the original Japanese version of Danganronpa V3 he used “Saihara-chan” to refer to Saihara. It was a nickname, and he used it on almost everyone, even the guys, like how he called Kaito “Momota-chan”. And “chan” is not limited to only girls, it’s just most commonly used for girls because it’s seen as more feminine, but it can still be used for guys (most commonly as nicknames).
