ON p53, I recall a presentation from years ago in which exercise was shown to upregulate p%3 and upregulated p53 was correlated with reduced prostate cancer. My understanding is that elephants have more copies of p53 and this explains why, even though they are very large and have more cells subject to cancer, they have less cancer than humans.

Nope. Sinclair main idea is that loss of epigenetic information drives aging. Gudkov proposes that it is DNA damage that drives aging. But they are entangled. Also one of the corner stones of the Gudkov's view on aging is degradation of immune system, which in turn explains accumulation of senescent cells. Sinclair says nothing about that. Sinclair's current approach is to reprogram aged cells' epigenome. Gudkov's approach is to stimulate immunity and suppress internal sources of DNA damage - transposones. Both researchers came to their views based on their own completely independent research paths.
