People have disputed a lot of this, due to how Koyasu Takehito expressed the character even back in Xrd. Koyasu says that the emotions are something Zato has to "relearn", similar to how Gears and Youkai "gain emotions" a little at a time. Izuna had a similar situation, where he didn't know how to emote when he was born.

Actually, it seems that they met before they came to Japan. Most likely Slayer met Nago during the expansion of the Ottoman Empire towards Africa and while Portugal was discovering trade routes around the continent. According to the storyline, GGStrive is around 2187. Minus 683 years, we would have the year 1504, 39 years before the first Portuguese arrived at Japan. They traveled together for a few decades and then traveled to Japan with the Portuguese, either together or each on their own. If the theory is true, that Nago is Yasuke, he most likely arrived around 1569.

Slayer's arcade end up being the best for me. Similar to Johnny's arcade, not only revealed Nago's backstory, but also more of Slayer's. We got that he's the second character in GG to exist because Nago was his first student in 683 years ago (also makes Nago to be the 3rd), currently Raven has the title of the oldest GG character. If they put Happy Chaos as the final boss instead, we would have a CINEMA arcade due to the lore bomb that is waiting, but it seems the Dev team decided to save the Slayer-HC interaction for the future lore coming up (maybe for a 2nd another story?). It's a nice pick to use Zato instead. With that, Slayer closes the Season 3 greatly, no, more than, it's a PEAK Season Finisher.
