2024 Introduction to Biophysical Techniques
The LMB Biophysics Facility houses a wide range of state-of-the-art and in-house built instruments that enable the molecular characterisation of macromolecular size, shape, mass, stability, binding affinity, dynamics and interaction kinetics.
The facility is primarily intended for hands-on research by students, postdocs, and group leaders, who prepare samples and make measurements. Biophysics supports users in advice on experimental techniques and results, training and in collaborative research.
In this video Chris explains physical principles that underlie light scattering measurements. He describes in depth how size-exclusion chromatography coupled with multi-angle light scattering (SEC-MALS) is used to determine molar mass in solution, using examples of experiments preformed in the LMB using proteins, complexes and detergent micelles. Additionally, he shows how conjugate analysis can determine the extent of modification of proteins such as glycosylation. Finally, it is shown how comparison of different light-scattering signals can give information on protein shape.
The Biophysics Facility is headed by Stephen McLaughlin, supported by Chris Johnson and Chris Batters.
