I have a question regarding the photolysis of water in photosynthesis and the release of protons. As I understand it, the Oxygen-Evolving Complex (OEC) goes through five states (S0 to S4), and during the transitions from S0 to S4, a proton is released at each state, totaling 4 protons. At the S4 state, two water molecules are split into oxygen and protons, releasing another 4 protons. Now, my question is: are the protons released during the transitions from S0 to S4 different from the protons released when the water is split at S4, or do we combine both sets of protons to get the total number of protons released during photolysis? If we combine them, it would give 8 protons released during photolysis, but I’m unsure if this is correct, considering that we often state 12 protons are released overall in the light reactions. Additionally, during the Q cycle, a total of 16 protons are transported into the thylakoid lumen, which should theoretically generate 4 ATPs. However, we usually say that only 12 protons are involved in ATP synthesis. How does this discrepancy work? Could you clarify this for me?
