Interestingly, that vowel sound can actually be made on a synthesizer or any sound really. You take an EQ and add 2+ strong peaks and move those peaks closer and away from each other the same way our vocal chords move as we manipulate air pressure in order to create sounds with our respiratory system. So basically you can emulate human vocal chords using an EQ or a filter on basically any sound. Not sure if that is happening here but I wouldn't be surprised given all the other synthesizers being used in this game's soundtrack.

For anyone wondering how this is made, it's simply 1 sample loaded into a sampler and being played at different pitches. Prolly to save resources since the arcade boards and SNES were very limited on memory at the time. Many of the sound effects are recycled IN songs in this game.
