I have mthfr c677t hetro, cbs, comt, and vdr snp's. I extensively consume activated B vitamins esp b1, b3, b5, b6, b9, b12 and pair that with glycine, taurine, EAAs, zinc, molybdenum, magnesium & vit C. The only folate that suits me is metafolin. The only b6 that suits me is P5P. Love niacinamide & nicotinic acid as well. Been ok and improved my health significantly. I do Not tolerate inactive forms of Bs well, and I also do Not tolerate methionine, cysteine, alpha lipoic acid well. Initialy i also had vit D intolerance, which I fixed by reducing D3 intake (500iu daily), while pairing it with B6, zinc, EAAs, magnesium. My favorite relaxation supp is pharma grade Glycine and favourite antioxidant is rutin/quercetin.

Dr. Courtney Snyder has a video on Overmethylation that's good and Dr. Janelle Sinclair's channel is great, great website with downloadable pdf's on supplement recommendations. I'm homozygous for MTHFR, COMT, VDR, MAO-A, SHMT1, and others. She recommends non-methylated B vitamins if COMT & MTHFR or homozygous COMT. I ordered the Seeking Health brand's Multivitamin One MF (methyl free) and get it tomorrow. I've been taking TMG for a few days with my Multivitamin that has methyl B's and do feel a little happier. TMG gets rid of high homocysteine, which causes inflammation and mental problems. It's recommended to use glycine instead of niacin to get rid of extra methyl groups too since too much Niacin can deplete methyl groups too much but too much glycine won't. Maybe this is helpful for you. This stuff is so confusing when you have lots of variants! Have you found any other good resources?

This is an excellent plan and I have used such with hundreds of patients. I now tend to use less of these the more organ meats and less lectins and oxalates I recommend. Good job!! It took me years to figure all that out and you did it!!

Excellent summary thanks. I have done this work you cite following Amy Yasko and many others, and over time as I applied more keto ot carnivore diets I discovered that these SNPS respond well to a proper human diet with less carbs and seed oils and lot of symptoms and lab imbalances just cleared up. The SNPs are still there, but their activity becomes balanced by the diet very often. I think supplementation can really help, and your advice is exactly what I did and do, and after struggling to stabilize these patients, we found they resolved naturally with ketovore-PHD type diets. All including me still need to watch our diets or we get symptoms, but it reduced the amount of supplements and symptoms so much. thank you!!!!

I've got both mthfr snp. C677 @a12. Does that over ride needing to look at all the SNP's? I take creatine & a grerns powder... Just feel generally ok. Added: higher dose b1&b3,b6,b12 a few times a week. (Noticed positive effects in mood, reduced anxiety, better energy. When b adding vitamins.

Walshintitute.org has a broader opinion on SNPs that is interesting, I do not think MTHFR trumps other SNPs. See Amy Yasko Genetic Bypass book also.

Check out the first part of the strategy of inyerpretation that Amy Yasko uses on her knowyourgenetics.com site. She offers free NRI analysis of some SNPs with a prioritization I find helpful. Her first step is to look at COMT and VDR together and decide what form of B12 and trace minerals are needed, especially lithium orotate, and then later folate is most optimal. Some people dont tolerate methyl B12 as well as hydroxy or adenosyl B12 and it can make a difference in some people. Later she gets into other combinations of SNPs and supplements in her free book Genetic Bypass and she has 4 or 5 other free books floating around online.

Frankly no. Li orotate is a super safe OTC supplement when taken at label serving ranges with a good track record for decades in many countries. You may be thinking of prescription lithium salts such as lithium carbonate and others that have more potential for adverse effects such as tardive dyskinesia.
