First use low polarity to high polarity solvent. So that some polar and nonpolar compounds also will come out.

For example in TLC, we can use toluene, ethylacetate, formic acid methanol in appropriate ratio. Then we can adjust the ratio of solvent, on second time based on the separated compounds

Thank you for your good explanation, at the explanation time you ask some amino acid Rf value but alkaloids, flavonoids , glycosides polyphenols and etc, this all phytochemicals are contain standard Rf value is available.if present kindly inform me.

Thank you for your response. Happy this video helped you. As like amino acids every phytochemical constituent have unique standard rf value. For eg, Compound like rutin and quercetin comes under flavanoids have a std rf value of 0.53 and 0.97. Like this every single compound contain standard rf value which will be used for the identification of the compound from the sample we're analysing.
