If you've wandered down a supplement aisle recently, you may have come across a bottle for sale that has "anti-aging" ingredients to help your body make a molecule called NAD+, better known as nicotinamide adenine dinucleotide. Your body definitely does need NAD+ to keep ticking, but what does the research actually say about taking supplements to make more of it?

And surprise surprise, exercise increases the amount of NAD+ in your body. Not saying a supplement is a bad idea, just that you can get more NAD+ by exercising, and not just NAD+ but a TON of other beneficial adaptations that increase both healthspan AND lifespan.

@@KokoHekmatyarr exercising is boring and monotonous for me too. But you don't need to do an entire routine an hour long 6 times a week, you just need 30 mins of high intensity exercise per week for all of the health benefits (according to some research)

I was taking NMN supplements before the FDA pulled it from the market because someone was trying to get it approved as a prescription drug (huh?). I was primarily taking it for the promise of increased energy (I'm autistic and also suffer from chronic fatigue, which may or may not be related to my autism). I saw a significant increase in both my general energy and my ability to be more productive while I was taking it. Ever since the whole FDA thing, any NMN supplements I have been able to find online don't do anything (meaning they don't contain any actual NMN). I don't care if it's a prescription or over-the-counter thing, I really want it back on the market just to help me boost my energy again.

But I say, if humans can live significantly long, I worry that there would be a suppression of the youth by the elderly majority. It is for the youth to inherit the world, the elderly should have no authority in dictating that. It is through the youth how society changes, it has never been done by middle aged or the elderly.

@@dh8203 This. I think it's a defense that or people feel they've had a pretty decent life and are okay with it ending in a short amount of time. Me, hell no. I'm sticking around as long as I can unless I actually hit a point where I'm truly okay with not being around. I see age as kind of a zombie disease. Everyone has it and every culture tried to make peace with it and many medical/scientific people tried to stop it.

I do very much agree with one thing in this video. Until they can prevent most if not all the potential horrible side effects of aging, modern medicine should be focused less on increasing life span and more on increasing "healthspan" as bro put it. Why would I want to live to 80, 90, 120, or any other number you can pick if chances are higher and higher that I could be a senile old invalid wearing diapers that someone else has to change for me? No thanks.

Almost certainly any medicine which extends the lifespan also expands the healthspan. Once you've reached the end of your healthspan, you're living on borrowed time and will die sooner rather than later.

Those health issues are frequently what kills old people. So if you're working on extending life, you're at least indirectly working on extending health.

Cycstic fibrosis was still a death sentence for children in the UK/Europe until the late 1960s!! We didn't get a treatment that wasn't tipping people upside down or hotboxing them until the 90s!! We've made so much progress in treatment of so many things that improve child mortality rates!! Look at us go!!

Food sources of NAD+: whole grains, mushrooms, vegetables, fruit, cow milk and products. For omnivores: fish, meat, poultry. Good food - tastier than pills.

I have been taking NMN since 2020 despite having stage 4 breast cancer. It is a slow-growing, estrogen-positive cancer that was diagnosed over 30 years ago. So far, it has been found in 7 different sites, throughout the years, so I've had to take a lot of toxic medications during that time. I don't know if the NMN is affecting the cancer in any way, but I rarely have to take any OTC pain medication, and when I do, it is for a spinal injury from an auto accident.

Keto diets might be worth researching, supposedly it can starve some cancers of the carb type fuel they need while the body lives of the ketone fuel At the very least, might provide an interesting read

What form did she take? I tried NMN for months (I think 1 gram/day), and it only gave me a very, very tiny bit more energy. And was very expensive.

NAD precursors do not make you younger, they increase NAD+ which gives you more energy, but they can be detrimental as they allow for the build-up of Nicotinamide within the cells, which is why they should be taken with TMG. The root issue is the steady increase in CD38, which eats into our endogenous supply of NAD as we age. Keeping CD38 in check is what is really needed, not more NR/NMN=>NAD.

Probably but there's a far cooler recent development in cat longevity. Cats usually die far sooner than their DNA would suggest they should live, because their kidneys give out. Recently we've developed a potential solution to the most common causes of cat kidney failure, which could potentially make cats live twice as long on average and reach 30 years old regularly!

You and me both my friend. Mine happened 25 years ago (in 4 days). I have two Herrington rods. But that doesn’t mean I can’t benefit from these supplements now. Especially with all the inflammation that comes from broken bodies. Even once they’ve healed.

D3 receptor polymorphisms might be addressed with Liposomal D3/K2, loss of melatonin, near zero by age 70, loss of CoQ10 with statins and age, loss of Glutathione, near zero by age 70, Mg deficiendy, D3 deficiency. Taurine has been shown to extend healthspan by 23%.

The mention of healthspan was poorly timed. You implied that NAD+ supplements would extend your lifespan, while classic methods like exercise would increase your health span. This is a common misconception around anti-aging therapies. In reality, an all anti-aging techniques specifically aim to increase healthspan, and lifespan increases as a result of that.

Exercise is CRUCIAL though because it is the only thing that can keep you out of a wheelchair in your later years. My 80 year old mother in law is still independent, mobile, and looks closer to 60. This is partially because she starts off her morning with body weight calisthenics. She can still pull off a six mile hike without complaint. The moment she stops trying to move is the moment she'll keel over and die, I suspect.

I don’t know if these supplements will keep me young forever, but they definitely have one very concrete benefit. They make my hair significantly thicker, something I didn’t expect. The change was noticeable enough that my barber noticed. After a couple months taking NR, she asked what I was doing because she noticed my hair was getting thicker. The downside, the effects don’t last if I stop taking the supplement. Given this, I switched to the much cheaper NMN. I’m 55yo, have perfect cholesterol levels and zero heart disease. Not sure if supplements are contributors or not. I eat a plant based diet and regularly exercise. Still, I’m about 10 to 15 pounds over my ideal weight, soooo … maybe. In terms of looking younger or having more energy, did not notice any relevant changes other than thicker hair. Truthfully, light therapy helps much, much better when it comes to looking younger. Just bought one of those light masks and, damn. They really do work … and work fast!

Nad+ is a power molecule that helps makes ATP in the mitochondria, going from NADH goes to NAD+ and is the biggest contributer to atp production, fats create huge amounts of NADH. The citric acid cycle and the ATP production in the mitochondria especially the f protein, the smallest molecule machine is so awesome

Strength training can help fend off some affects of old age, doesnt even need to be hypertrophy. Exercising in general helps.

I consumed NR TruNiagen daily for 2 years with no other supplements. It did make me feel alright, however, over time it caused Peripheral neuropathy numbness in multiple finger tips. As I have stopped for a few years the numbness only remains slightly in one finger tip and hopefully will recover over time. It also caused knee pain after exercise. I no longer suffer from this knee pain issue after I stopped consuming the product. I’m a male in my mid 30’s. Buyer BEWARE !!!

I didn't get neuropathy with NR but I did get vertigo and nausea - so I only had about ten or twelve pills. There are so many drinks and foods that have added niacinamide/nicotinamide like "Vitamin Water" that it is quite easy for people to take too much and get nerve damage. Especially if they take supplements as well. It is well known that overdoses are dangerous. It is usually mentioned in the tiny small print on bottles and packaging that nobody would think they need to read because - vitamins are healthy, right?

Thats exactly they try to do that tho. If your mitochondria and immune system works properly you can avoid getting cancer and antiinflamatory mechanisms really important in that.

My memory and energy levels really took a nose dive in my early 40's and heard about nmn tried it, and I had a ton of energy my memory/recall is way better and I have way better reactions. I know people that tried it and said nothing seemed to happen so maybe my body didn't make enough? Idk but it has been a miracle for me, and at a pretty low dose.

NR and NMN are typically expensive and the market is full of fakes. For some reason, people don't seem to realise that NMN and NR are just different forms of vitamin b3 - and you can therefore get a NAD booster by just buying niacin (b3) supplements, which are way cheaper. I do not fear death, but I would love to live as long as possible. The reason for this is to experience as many incredible achievements in science as possible. I am curious what AI will be like in the future, how advanced medicine will be, and how far humanity will travel into the cosmos. When I do pass away, I would love to donate my body to science because I would die peacefully knowing that I am making a contribution in science even during death.

I use NR, but ... NAD+ going down may be more a design feature rather than a bug. Seems to be related to inflammation and senescent cells... You don't want to rev up the engine when the car is starting to get rickety.

For me (not Asian) it's wearing sunblock consistently that has protected my face from the signs of aging. There's no such thing as a healthy tan, no matter what skin tone you're starting with.

No. Your body produces enough of the stuff when you are young. A deficit only occurs as you speed past 60. 😂

Yes and no, mostly no. If you want to preserve youthful appearance, do each step as I tell you, some is easy, some is harder *but you should do all* - Quality skin care routine (cleanser, toner, chemical exfoliant, serum/active, moisturiser) done daily, with a different routine in the AM and PM, followed by a sunscreen designed for the face (non comedogenic) ALWAYS FOLLOWING AM ROUTINE [Side note: consult a dermatologist for your personal compatibility but alternating aha and bha in the AM /PM with a QUALITY formulation will do wonders] - From age 23 onwards begin exploring a prescription for retinoic acid from a dermatologist - following this you must *never* forget to apply sunscreen - no smoking, vaping. Edibles are okay however - minimal drinking - exercise (4 hours of high quality intense exercise every WEEK) - high quality balanced diet ---------------- In the event you decide to take nad+, understand that nad isn't the silver bullet it's been made out to be, *the key issue is cd38 regulation* and while you'll hear you can eat various herbs, like dried parsley. It's long-term wear on kidneys isn't worth the extremely weak and broadly unreliable results. A better way to think about this problem isn't ageing = bad, instead think, youth = a set of behaviours and tendencies. For example, youth do not drink, nor do they smoke, they exercise regularly, engage in meaningful social connection and have a strong self care drive - *start here* and then add the skincare and drugs

This doesn't make any sense. Anti-aging drugs prevent cell and genetic breakdown, which has nothing to do with normal growth from childhood to adulthood. These drugs only do something when you're done growing and your body starts breaking down (that's what they mean by "aging").

If you're talking about kids with progeria, that would probably not really translate into results for "baseline" humans, even if it did help the kids with it. Progeria specifically is the result of a genetic mutation that disrupts nuclear integrity, which has a lot of (apparently) downstream effects that appear very similar to aging. But this should not be confused with the actual aging process. Since actual aging seems to be a combination of telomere shortening, build up of oxidative stress, accumulated mitochondiral dysfunction, genomic damage, epigenetic changes, and cellular senescence (likely in response to much of the above), anything that might be a silver bullet for progeria would probably have little-to-no positive effect on people without progeria.

I was taking NMN for chronic fatigue. I definirely felt like working out more and had better muscle tone and had a bit more energy. I djd feel a little more anxious too, bur unlike many supplemebts I didn't feel like it was an effect I couldn't chalk up to placebo

1. The largest killers of humans(not to mention a large portion of suffering) is by far heart disease, Cancers, diabetes, dementia, all accompanied by frailty. An all too small percentage of the federal budget is allocated towards it. This needs to change. 2. My genuine question is how many people who turn 80, but in perfect health(body of a 25 year old) would be OK with simply saying "that was enough life."? How many would choose to continue living and exploring and learning.

I understand your concern; it's a common one, but: 1. Birth rates are falling 2. Birthrates that are kept afloat by immigrants, are temporary; Mexico is at ~1.7, similar the US, according to the PRB, as an example.

I suffer from a disease that causes chronic inflammation and take some prescription meds for it, as well as some doctor-recommended supplements. I also strictly adhere to a low histamine diet. If anything that seems scientifically promising can help with this awful disease, I'll try it and monitor the results closely. Hopefully, these NAD+ supplements are made in a high-quality control facility, with proper dosing, and are truthful about actually using the supplements they say they are using.

Polyunsaturated vegetable oils can be helpful for inflammation. Though the wrong ones may inflame more. Hempseed oil seems to have the precursors for the prostaglandins that reduce inflammation. Not the high CBD type oils, just the vegetable oil for salads and mashed potatoes. Also tastes good.

yeah they really didn't talk about my favorite NAD+ precursor, namely niacin (and not the long-release kind). Yes it makes you beet red, and yes. you itch a bit, but taking 500mg one time per week really does work well with apigenin and TMG.

I've experimented with the precursors. It's also true that calorie restriction can extend life and health. As I contracted diabetes type 2 I went on Keto. A lot of good things started happening. It turns out the constant glucose metabolism most people have is not in our evolutionary adaptations. Neither is a high carbohydrate diet. A ketogenic fat metabolism has intriguing advantages. One is that oxidative phosphorylation creates more NAD+ instead of converting it to NADH. It's also anti inflammatory. To fully benefit from autophogy you need exercise and several 3 day fasting sessions a year. Just taking a NAD precursor is ignoring most of the metabolic puzzle.

Totally agree. I used to take nmn, but stopped when I started trying the 168 plus keto diet. It took me 18 months to lose around 20 pounds. Worked very well on my prediabetes as well as age reverse (20 years younger). No one will believe I am 59. People think I'm in my late thirties at most 😂

I tried NMN for a few months and didn't notice any effect, good or bad. I stopped taking them. I'm a one case example, but I wasn't impressed. I'm just going to cut sugar, eat whole foods, and exercise.

If you want to maximize NAD+ production, it seems to be more or less just as simple as "eat well and exercise" 😂 Obviously this includes whole foods with a tendency toward green vegetables, with a full spectrum of fatty acids from fish, red meat, nuts and seeds.
