How should I practice this at home? also is most of this piece a left hand lead or a right hand lead?

Practicing on a bar stools or set-up 4 round pieces of cardboard on a bed or table can substitute for the playing zones of timpani. Work on finding whatever stickings work best for you. Some people use crossovers and doubles more than others. It's a personal choice; just make sure you have a great, consistent and even sound across the drums. Best of luck on your audition!!
