Sunak was Prime Minster and leader of the Conservative Party. In the UK, the leader of the largest party in Parliament is the person that becomes Prime Minister. (The Prime Minister is the person in charge of the Government.) The Conservatives lost the recent election, i.e. Labour is now the largest party in Parliament, so their leader (Kier Starmer) will take over as Prime Minister. Technically this works by Sunak resigning and the King asking Starmer to be Prime Minister, because technically the Government is employed by the King. However, Sunak had no constitutional way of staying on as Prime Minister and if he had tried to the King would have essentially fired him. In the speech Sunak also announced that he would resign as leader of the Conservative Party, which he isn't technically forced to do but pretty much always happens when a party loses an election. If he did try to stay as leader of the Conservatives it's almost certain they would use the Party's internal rules to remove him and select a replacement.
