Because if Nezuko were to fight to take the uppermoon 6 spot, she would have to fight Gyutaro because he is the true uppermoon 6. They do not think she is fighting for uppermoon 6 rank because Nezuko would have to challenge Gyutaro and Daki for a blood battle. Where demons challenge for the demon’s ranks. Like when Akaza challenged Kokushibou three times but lost. Also the winner of the blood battle absorb their opponent. Kokushibou let Akaza live because he is useful. Also, the blood battles happen in the infinity castle and Muzan is present when it happens. Also, Muzan is just mad that a demon that has never eaten a human has so much power.

Typically, when a lower ranked demon (especially considering the circumstances that the Lower Moons have been disbanded) fights an Upper Moon, its to take that Upper Moon's place. So without the context of Nezuko being a demon who does not eat humans and just sees a demon fighting Daki, it would be considered a canon reaction for them to assume she's fighting for the Upper Moon 6 rank. Demons don't fight other demons unless they want to show dominance, fighting over food, or move up the hierarchy Muzan placed because they can't kill each other, only Muzan can. So I totally agree with you there. However, the problem would be that Muzan would not see it as an actual blood battle for an Upper Moon rank since Gyutaro's not in the picture. To Muzan, Gyutaro's the true Upper Moon 6 even though the siblings share the same position. So if Nezuko were to fight for the position, she'd be fighting both siblings or just Gyutaro. Muzan is likely mad because he sees Nezuko as a threat since she severed her mental and cellular ties (aka he can't see through her eyes/read her thoughts and all that stuff he can do with the other demons under his command) + is confirmed to be working with Tamayo and another demon he does not recognize but he still underestimates her. A rogue demon to him is a liability especially if they know Tamayo who's connection with him has been cut a long time ago, but he didn't expect Nezuko to be so powerful and have qualities like an Upper Moon AND the kicker is that she's not even under his command with abilities like that. So that ultimately moves her up as a threat.
