These methods have varied purity outputs so usually two or more of these methods are used in combination.

Isolation and purification are used interchangeably for a process where you obtain virus in a 'pure' form, free of cell extracts. Isolation is also used to signify cultivation of virus on a suitable medium.

I have mentioned it towards the end of ultrafiltration segment. Normally a pore size of 0.002-0.1 μ and MWCO of approximately 10,000 to 100,000 Da works.

They are used separately as the criteria for separation is contrasting in both these techniques.

Viral lysate refers to the extract of the virus that we are working with. For experiments where we need the whole virus, lysate is prepared by centrifugation of the viral culture and subsequent inactivation.
