When I discovered that in Sinclair's study, low resveratrol had the best longevity results, the first thing I did - was trying to get Dr. David Sinclair’s response. Did he answer to me? You’re about to find out.

Do you really believe Traditional medicine, is all about disease prevention not health optimization? Traditional Medicine is all about big Pharma and providing a pill when you are presenting with sickness and disease--too late IAW. Functional Medicine is about disease prevention and health optimization to the contrary. IMHO of course.

Also, remember DS has cancer in his family, and IIRC on at least one video he said he has inherited a gene for lung cancer so if he knows he has a higher risk of cancer and he also knows high resveratrol has a cancer inhibiting effect then that would probably influence his personal resveratrol dose.

Could be Dave, that would make sense. I couldn't find him mentioning that, but I could be a contributory factor. My research shows to separate benefits between low and high s take what fits the situation.

I also remember DS talking about his mom, who died in front of him, from lung cancer. And that made him turn into longevity research, quite a story. I think he said she was a smoker.

Unlikely because if this is the reason he is taking high-dose over low-dose he would have talked about it. DS just hasn't realised that low-dose and high-dose serve different functions. To him, higher is always better. What WM has discovered is very counter intuitive.

It is such a complicated compound! Besides the longevity factors that favor low dose, there are so many other effects. Increased nitric oxide production which has wide ranging cardio benefits and activation of the AMPK pathway are 2 primary ones. These are other benefits that may not move the needle on lifespan, but may affect healthspan.

I started back to take higher dose of resveratrol. This is because I think the effect is mostly on the senescent cells, which you want to neutralize/catabolize. It basically mimic an intermittent fasting of three to four days… this is likely why, opening mice on high dose, they found their organs were still young… or, at least, I hope

Sinclair believes that resveratrol and NMN work synergistically to extend lifespan. This is the reason why he probably does not care about taking a gram. In Sinclair's analogy, resveratrol is like the accelerator pedal of a car. It helps to activate the sirtuin genes, which then use NMN as fuel to do their work.

Great video course and very in detail, appreciate that. 🙂 Curious about one thing. Doesn´t anything that lowers inflammation interfere with exercise if taken within +/- 1 hour and supress some of the inflammation caused by the exercise thus yielding a lower result from the exercise…?

Thanks, this video confirmed for me my decision to not take resveratrol. There’s too much confusion and disagreement and the cycling requirement and exercise effects and chances to make mistakes make it too complicated and risky for me.

Great, very insightful series. I have reduced the dosage of resveratrol to what you are recommending. Off topic: are you by any chance planing to look into OPC, as it could have a similar effects like resveratrol, but there are also studies which show, that it could be a senolytic.

I heard that the body doesn’t absorb Resveratrol very well - not sure if the body absorbs the whole 1g that he is taking. Or the body only absorbs 300mg from 1g.

@@WellnessMessiah And that would just make everything more interesting for everyone else. But I think you are right that he would never agree to an interview with you. I have seen several interviews with him, and most is more around what Sinclair does and his reasoning for doing so, I haven't seen anyone do a deep dive into the data the same way you have. That would just lead to a much more intesting dialogue for him to actually have responses to some concrete data that doesn't really match what he is preaching.

Sinclair won't say anything. The stuff doesn't work but he sold his formula for millions. So if he would say now it doesn't work, it's a carrier ending move. You can ask him, but he will ignore you. Better not ask this and leave the possibility open to ask him something else in the future 😊

I feel like he didn't do in depth technical interview since Peter Attia, maybe I could be the one. Will have more time once my wife is more recovered from her stroke. How would you take it If I were to interview him without asking about NMN controversy? Feels a bit inauthentic on my end

Your hypothesis about two distinct mode of actions of hi and low dose resveratrol is certainly interesting. Have you seen any studies confirming it, by any chance? Also, if this is a case indeed, should we take high doses of resveratrol as well as fisetin and quercetin on cycling basis? Say, stick to the lower doses on the daily basis (after the exercise and with some fat and/or wine) and taking high doses once a month for a week or a few days? And two more thoughts if I may. I have had the worst experience with resveratrol specifically before I finally found a good source of it. Even so, how do we know how much of this compound is in mixed types of supplements, which became available and affordable on Amazon? If the bottle says 650mg of resveratrol in proprietary mix… perhaps, it contains only half of what they say it does in a single capsule? It very well might be that we are taking low dose even if they advertise it as a higher dose? Lastly, are there any studies on synergetic effects those three compounds have in each other? Should we take them all three at once or cycle each and every one independently? Thank you for your time and great content, colleague!

Metformin creates a B12 deficiency, and a B12 deficiency causes neuropathy. So it's more than likely diabetic neuropathy is caused by long term metformin use

How about rotating 1 or 2 high doses of resveratrol into your weekly regiment in-combination with the low doses ? that way you can activate all the pathways or is that not a thing ?

I feel like it would be very interesting to see someone take high dose resveratrol everyday for 6 months or so and do biological age test. Followed by 6 months of low dose at every other day. See what differences in aging occurred.

Yes, that's an excellent experiment. Honestly, I would go for a year of each, but always cycle. I would the Horvath clock test over other biological tests.

thank you, very good. towards the end you say, low dose R. works in *humans* for longevity. Mr Lustgarten, in his YT channel "conquer aging" which is about bio hacking aging, has an episode about R. ... He mentions by citing some papers, that R. is only extending the life of obese mice to normal, not above normal. what do you think? About "cycling"... I pretty much cycle everything, including aspects (micro + macro) in nutrition, types of sources, relative amounts, supplements, on a medium and on the shorter time scale.. I think, this keeps the adaptability of the metabolic system up.

I think not just cycling Resveratrol but cycling it’s doses would be a good way to go…it similarity with quercetin is uncanny so may be similar approach…. Thank you for all the interesting insights. There are so many other interesting supplements to take, I feel like Brian Johnson on some days, hope it all works out well long term - my blood work looks stellar so far. 😊🤞

Yes it makes sense AT. In fact, that is exactly what I would do over age 60 (said that on episode 3 I think). Right now, lacking above average cancer genes in may family, I think I will let long fasting kill/ eat renegade cells. Family history will convince me to cycle both doses sooner.

Trans-Resveratrol between 500 and 1000 mg serves the specific purpose of accelerating the conversion from free testosterone to DHT while also inhibiting the aromatase enzyme, besides helping the venous system (in synergy with liposomal vitamin C) from bleeding during excessive efforts, like when...you use vacuum devices for P.E. 😉 I guess that after this likely purpose will be reached, Sinclair himself will get back to the 250 mg dosage

The main problem of resveratrol is the poor solubility and high clearance. Sinclair know that very well. Talk to any Harvard and they will tell you how they feel about him. Also, it is not synergistic it is additive that’s the other problem. Synergy would be if they are on the same target increasing residence time of the compound on the protein. This is a pathway that he is looking at not a single protein.

Partially, yes. But only when burnt. Resveratrol works with meals. Oleic acid works during fasting, exercise, etc. That's why they are complementary.

Personally I think lower dosages coupled with synergistic molecules that work in different parts of the anti aging cascade is the optimal way for rejuvination. For example, Piperine seems to potentiate the benefits and absorption of most of these molecules, hence lower dosages could be used.

I am concerned about side effects. One is that it blunts recovery from exercise. I exercise a lot everyday. I’m trying to make progress not maintenance. but Sinclair is apparently too busy working to put in the time I do. He does hot and cold plunge and one or two workouts per week. I don’t want to take metformin either for that reason too.

Hi Joe I wouldn't be too worried about it if you take after exercise, 24 hours before intense session. Check episode 3, or consider re-watching it again for confidence. And the fact that you take a lower dose of resveratrol, it's lower than the studies, it's a double coverage.

It’s incredible how much scientist ms are learning about the human body but what I do know is that every few years they change their mind in the efficacy of drugs . What seems safe and effective now may be deemed unsafe in a few years . I wouldn’t jump in the bandwagon , pay huge amounts of money for unproven drugs and potentially subject yourself to harm in the process. You need longitudinal studies to see how safe they are

SInclair is a doctor, he developed it. If we don't know what is going on, especially if he knew that low portion is for longevity and high is for killing damaged cells then maybe it's about promoting Resveratrol and money 🙂 That's my conspiracy theory. Sinclair is to clever to be not aware of it. Besides he knows how to promote 'his' products. He knows market, how it works. If I had no interest in this I'd never mention how much I take, because it depends on so many aspects ! How he can do it being a professor ? Imagine that ! Awareness is strong attitude of what people do, because sometimes even we know something we treat it as a something normal. He shares information like suggestions/promotion of something (what many Y.Tubers do saying the opposite that it's is not an advice ofc.). Also looking at 4 influencers (are not doctors or dietician or any other kind even, Rimon, you are curious, you dig a lot to check information and that is very useful to discover many unusual things related) related to DNA brand, they claim that they don't work for them. This is so weird. So, strange 'coincidence' 🙂. Their channels are so different and still so similar. They never mention each other 🙂 That shows only how YT influencers work 🙂Recently one of them started taking 2gr of NMN saying it's for fungus. Many of them shows their stacks. These stacks are enormous ! I know one thing, this is anourmous amount of money as well around it. Be careful people. Don't lose control about what you need, especially if you are thin and healthy thinking you will increase drastically your life. Look for natural alternatives of NAD and Resveratrol !

@@WellnessMessiah maybe it was mould, I'm not sure, anyway 2g of NMN for is a bit too much whatever it is, I noticed that using 0.3 - 0.7 g is for me best (64kg, athletic). And even when I take more somehow I don't feel well (but this I have to double check why)

Brad Stanfield has done videos on resveratrol. He's stopped taking it because of lack of evidence of benefit and evidence of harm. Sinclair has made a lot of money off it, and knows the updated evidence.

resveratrol is rapidly cleared from the body. I think low dose perhaps twice a day might be more reasonable than high dose.

Maybe DS is making an educated guess that if resveratrol seems to have a positive effect on chronic or metabolic issues at high doses, logically it would extend life of those affected by those diseases which seem to be at epidemic levels in humans. But that should also be observed in mice too. If a mouse is fed a terrible diet or made to have cancer, can high dose reseveratrol extend their life vs low dose ?

Excellent comment Bob. The studies in mice with high dose resveratrol show a reduction in tumor, which SHOULD be translated to a longer life. I mean, I cannot say how reducing the most dangerous disease on this planet won't extent. But I don't remembering carrying the experiment for 2-3 years to see.

Mine is only 100mg resveratrol. Is this worth even taken or should I increase.very hard decisions as everyone including Sinclair seem to contradict themselves. .

I take sometimes 50 or 100. I'm pretty sure about this dose as long as after exercise, not before. We have enough data, resveratrol is terrific and rare in food in good doses unlike otherlongeivty molecules.

So, I think he is making a mistake to just write off Resveratrol completely.

According to my knowledge, Vitamin C is a potent antioxidant and will not cause more oxidative damage to our body even at high dose usage. This is because the metabolites of vitamin C are not pro-oxidant, while that of the antioxidants like Vitamin E are. And that might at least partially explain the research outcome Rimon mentioned in one of his previous video about resveratrol: Supplements of antioxidants like Vitamin E actually increase mortality, while vitamin c does not.

I watched David Sinclair tell Andrew Huberman that he takes anywhere from half a gram to one gram of resveratrol per day. I also saw Sinclair tell Joe Rogan (in a different video) that he takes a half a gram of resveratrol per day. And of course in his Lifespan podcast he claims to take one gram of resveratrol per day. So, either the dose isn't that important to him, or the dose is a work in progress.

I bet he just measured either his bloodwork and trial and error'd the dosage for optimal values, or just goes by resveratrol concentration in his blood to reach enough for activation of the targets he wants.

Awesome part is it's safe to take 3G a day for a bit, so I'm taking 3G for a month with other synolitics, then dropping the dose to 500mg moving forward. If it does nothing. Great. If it does something, maybe I clean house.

Resveratrol may have synergistic effect with many substances. I would expect Betberine to be on that list as well, because of its likeness to Metformin…and some more

@AT-xl9db maybe, but leucine being one of the building blocks of cell and muscle growth at a molecular level is said to have much more importance than just a distant synergistic effect on cell growth and repair

Thanks to your episode 2, I stopped taking high dose resveratrol shortly after I started and now take 100 mg every other day. What do you think of this. kzbin.info/www/bejne/fKvPaXlqjLaZitU I take L-Leucine midday with other longevity supplements not just resveratrol but NMN 250 mg, the recommended salvage pathway R-alpha lipoic acid, TMG and apigenin, AKG 1000 mg. I'm thinking of buying the Sirtuin 6 activator. What is your opinion? I take many other supplements at other times of the day. For building muscle I take at night BCAA and HMB. My muscles have grown huge and strong despite doing no exercise. I'm on OMAD 6 hours on, 18 hours off, mostly keto but not all keto. Also I'm aware of resistant starch and refrigerate any and all of it. It seems to work and got the okay from my doctor. I'm 74 years old and feel like 30.

Resveratrol activates AMPK. AMPK is a double edged sword. Prior to the establishment of cancer, AMPK acts as a tumor suppressor. However, following the onset of cancer, AMPK has been shown to either suppress or promote cancer, depending on cell type or state. This is because although it inhibits mTORC1, which facilitates cell proliferation (cell division), it also activates mTORC2 and cell survival.

Well clearly 1g is safe, Sinclair does it, and the superiority of low vs high dose is hardly statistically significant, in fact many studies show a steady improvement in hypoglycemic effects with higher doses. Since the blood sugar is king when it comes to longevity effect, I am sticking w Sinclair on dose

The research on resveratrol was manipulated; it doesn't work. Dr Sinclair does not take ANY resveratrol, never mind so much. He has a vested interest (money) to keep the act going.
