So question: In regards to somebody like Villager or Isabelle why can you still repeatedly alter their course with their balloons and it count but you cannot allow Jigglypuff or Kirby to use multiple inflates? You still need to hit the button multiple times for them to increase their height with the balloons

The rule is one move one time, not one button press. Thus, jumping multiple times is not allowed. Pressing buttons to move Villager’s balloon up is part of Villager’s up special. However, if you prefer, you can just hold the B button and he’ll go up as well.

The rule isn’t worded the best and definitely sounds like it would allow that. Prowder just meant things like Shulk up special swinging twice and stuff like that. In the case of Ness and Lucas, they are using up special twice, so it isn’t allowed. It’s like if Sora did side special after up special (not allowed).
