Nucleic acids have an absorbance maximum at 260 nanometers and Proteins have an absorbance maximum at 280 nanometers. The 260/280 ratio is calculated by dividing the absorbance at 260 nanometers by the absorbance at 280 nanometers. A ratio of about 1.8 is generally considered pure for DNA.

Yes but the pH of the salt varies with it for plasmid we generally use acidic pH but for genomic DNA the pH is close to neutral
