Apoptosis is a type of cell death, which is characterized by an orchestrated regulation of the disassembly of cell organelles, that is, it is a programmed cell death. This death is present in some types of damage, where cell viability is compromised: genetic mutations, or physiologically, to control cell number. There are two common mechanisms by which apoptosis occurs: one is the mitochondrial (intrinsic) pathway, where everything arises when the anti-apoptotic proteins (BCL2 and BCLXL) stop working, when the initiators of apoptosis (BAD, BIM, Puma or Noxa) send signals to the pro-apoptotic molecules (BAX and BAK), increasing the permeability of the mitochondrial membrane. The other mechanism is the extrinsic pathway, characterized by the binding and activation of the TNF-1 receptor, triggering in both pathways the activation of caspases (9 for the mitochondrial pathway and 8 or 10 for the extrinsic pathway), both pathways converge to activate other caspases (3 and 6). In addition to these two pathways, there are others that have been discovered somewhat recently, necroptosis, which has similar characteristics to necrosis and apoptosis; pyroptosis, which forms the inflammasome, triggering the classic symptoms of an infection or inflammation; and ferroptosis, caused by the accumulation of ROS and iron in the blood. All this triggers the release of nuclear fragments into the cell cytoplasm, to be finally digested by phagocytes. It is important to consider that apoptosis differs from necrosis due to mitochondrial damage. Finally, in the microscopic form, apoptosis can be visualized with reduction of cell size, chromatin condensation, vesicle formation and presence of macrophages.
