Thinking that when the birds fly it makes the truck lighter is like thinking when the fish swim to the top of their aquarium it makes the tank lighter. And as the video pointed out, it is in the open air, the pressure is being dissipated and distributed over a big area. In the truck, there are walls and roof which holds the pressure. If you, however, remove the walls and the roof, then sure - flying birds will reduce the total weight

In the grated for scenario, the pressure would still dissipate, from the height of the truck bed, to the road(which tends to be reasonably high). Although I wonder if there would be a significant effect from vacuum created by the birds pushing air down with roof and walls intact.

If the truck had ample ventilation(which a truck carrying live cargo definitely would), then the fluid would flow out of it with the added air pressure of flapping wings. Maybe the trucker did not think the problem through but VSAUCE definitely did not. That's the problem with theoretical physics. There are often variables that a theory does not account for.

I don’t think so. Even if the ground of the truck was wire, the force of air would still have to end up somewhere- aka, the bridge below the truck. It may fare a bit better since the force would be more spread out rather than instilled via low surface area wheels, but the birds still have weight that they are forcing onto the air.
