you can assign randomly but they must be according to orbital (s,d,p). They indicate sign of wavae function (no significance)

because we are considering z axis as internuclear, so they're perpendicular to the internuclear axis and will form pi bonds.

If you are talking about the 16:03 , even the plane passing through the nucleus( perpendicular to the internuclear axis) will be a node, because its a p orbital. Sir only considered the plane between the nucleus to explain the concept
@@Riya_draws If you rotate it by 180 degrees, you should get the same shape with the same signs. If you rotate that orbital diagram, you'll get opposite signs at the same positions make it cylindrically unsymmetric

it is a line originating from symmetric centre of structure and extending outwards dividing the structure symmetrically. You can check sir's lecture of shape of orbitals.
