1.- Sukuna and Kenjaku are from the same era, we don't really know if they were friends or enemies or acquaintances. 2.- Yes, Sukuna ate his brother in the womb and yes, that brother's soul ended up in Kenjaku's hands, we don't know how or when but we know why: Kenjaku was obsessed with Sukuna and wanted to create his own Sukuna. 3.- Kenjaku keep an eye on Sukuna's lineage and when the time was right (we don't know what the conditions were) he took Sukuna's brother soul and implanted it into the baby that would eventually become Yuuji's father. 4.- Kenjaku supplanted Kajori (most probably kill her too) and using his body manipulation techniques he created Yuuji inside of him and manipulated his DNA to be the perfect Sukuna vessel, this because he wanted to use Sukuna's brother soul as a catalyst to create his own Sukuna. 5.- Something went wrong and Yuuji just got his father's soul embedded, this process killed his father and Kenjaku (deeming the experiment a failure, or having finishing the first step of his plan) staged Kajori's death and hooped into a new body.
