Hello, can this autoclave be used at a temperature of 250 degrees Celsius? I also need to make an autoclave for my university project. Of course, the autoclave environment should be at 250 degrees Celsius for 3 hours. Do you have any comments or suggestions?

How do I operate this DIY Autoclave to sterilize a liquid?

In order to sterilize, nothing needs to be redone. With increasing pressure, the boiling point increases.

sterilize a bottle and it's cap (not sealed) in the autoclave. Add or mix in your liquid, such as liquid media, sterilize the bottle with the cap just started (1/2 turn, if its sealed the pressure inside the bottle can cause it to explode), if there is no cap, place aluminum foil over the top. It helps to pre-warm the liquid to about 60°C before starting. It should be sterilized for about 30-45 minutes at 115°C or 20-30 minutes at 121°C. When done, seal lid or foil top, using oven mits remove the bottle and place on countertop to cool. If you need to use the media before it comes to room temperature (such as pouring plates), monitor it's cooling and use when appropriate.

It also helps if you have a container for the media, a box to sit it in, that way any spilling or boil over is caught instead of dirtying the autoclave. Any #5 plastic should do the job, polypropylene. It's often the hard black plastic used in takeout boxes, the same kind that is microwave safe.
