Analytical Instrumentation - Fluorescence & Phosphorescence measurement
----------------------------------------------------------------------------------------------------------
You may have come across terms like fluorescence and phosphorescence. You may have a lot of questions in your head regarding this topic like: What does fluorescent mean?, How does fluorescence works?, How does fluorescence spectroscopy work?, How does fluorescent occur? You will find answers to all these questions in this video.
In the following video, viewers will learn about the basic principle of fluorescence and phosphorescence. The generation of fluorescent and phosphorescent emissions is discussed in detailed using an energy level diagram of a molecule. In the energy level diagram we can see different levels of molecular energies. The electrons when in pair are either in singlet state or triplet state. The basic definitions of singlet state and triplet state is covered and pairing of electrons and their spins is explained in the animation. Once we understand the energy levels corresponding to singlet and triplet state, it becomes quite clear to understand fluorescence and phosphorescence. The fluorescence and phosphorescence emitted by a sample under test is used to perform qualitative and quantitative analysis on the analyte. It is performed using spectrofluorimeter. The basic principle of measurement of fluorescence is also explained in the video.
At the end of the video, viewer will also have a clear idea between the differences in fluorescence and phosphorescence and will be able to compare the two topics.
Hope you like and enjoy the video. Give us a thumbs up. Stay calm and keep learning. Subscribe to the channel to learn about such more interesting concepts and to never miss any new updates.

I appreciate the efforts of making this video with intention to simplify the concept. However, i noticed that you have forgotten to mention several things including the internal conversion, types of molecules that can fluoresce etc. Another thing is that you have wrongly mentioned the lifetime in the singlet excited state as varying from 10^-8 to 10^-4 seconds. These l lifetimes are I few nano seconds only.

The Jablonsky diagram doesn't depict orbital energies, but the energy states of the molecular electronic system as a whole. You cannot put electrons on this diagram like you would in a molecular orbital diagram. Basically, every energy level in a Jablonsky Diagram represent its own personal molecular orbital diagram which is omitted.

@lokeshgnanasekar
@lokeshgnanasekar 2 жыл бұрын
Acetone may fluoresce when a high intensity monochromatic sources are made to fall incidentally. It is difficult to analyse whether all compounds fluoresce or not, since we consider that phenomena generally in the visible spectrum; What if it emits in invisible wavelengths...Hope you know that's why we use UV/Vis spectra, thought many paper suggests they can identify, it has many limitations Edit: For vibrational energy we use IR
