here is a question! if the methylation is saving the prokaryotic DNA to be cut be by the restriction endonuclease, how can we cut some DNA of interest of a prokaryote by a restriction enyzme? Will the enzyme be able to cut the DNA of interest in the presence of methylation?

Sry for interruptions 😊... As I know... Vectors have multiple restriction sites but an endonuclease is confined to cut the chromosomal DNA only at a specific site...( like either blunt end or a staggered cut) ... Coz for this unifying feature , a particular endonuclease is allowed to cut both the gene of interest and vector...

Mam u said that ligases are needed to make phosphodiester bonds....Then why alkaline phosphatase is used to avoid bond formation......If we dint provide ligases ,they will remain circular ..... Correct me If i am wrong... Thank you

Your lectures are very clear , easily understandable and without scribbling anywhere on the white board. After the end of the lecture if one sees the white board it your lecture when read should be followed. Sorry to point out one flaw when you draw a straight line below the Binomial nomenclature of any plant or animal or prokaryotic bacteria.Separate line should have been drawn below the generic name and a separate line for a specific epithet. I really enjoyed listening to your lectures.
