here is a question! if the methylation is saving the prokaryotic DNA to be cut be by the restriction endonuclease, how can we cut some DNA of interest of a prokaryote by a restriction enyzme? Will the enzyme be able to cut the DNA of interest in the presence of methylation?

1) Mam can alkaline phoshphate enzyme remove OH also from the genetic material or does it remove poshphate only? 2) does methyl group gets attached to the N2 bases of host's genetic material only?

Because MS can recognize its cells DNA strind( bachterial ).it works for endonuclease by diffrentiat its cells dna by methylates to it. Thats how endonuclease cuts foreign DNA .

Does each endonuclease have a unique property of cutting at different sites and how does this done? Kindly clarify my doubt, ma'am.

Sry for interruptions 😊... As I know... Vectors have multiple restriction sites but an endonuclease is confined to cut the chromosomal DNA only at a specific site...( like either blunt end or a staggered cut) ... Coz for this unifying feature , a particular endonuclease is allowed to cut both the gene of interest and vector...

In NCERT, it is written that the Roman numbers indicate the order in which the enzymes were "Isolated". Please check.

Mam is gonna wrong in that case there is hydrogen bond between c and g which is cutter while the line shows phosphodiester bond

@@MuhammadYaseen-dd1uw NO... as hydrogen bonds are present between two strands of dna ..they are not broken...because..here we are seperating the G and C ...which are present complementarily in the opposite strands...as the dna is seperating at the same place in both the strands...hydrogen bonds need not to be broken...

Mam u said that ligases are needed to make phosphodiester bonds....Then why alkaline phosphatase is used to avoid bond formation......If we dint provide ligases ,they will remain circular ..... Correct me If i am wrong... Thank you

Prakash Raj Bro actually we need phosphodiester to protect its own joining again but whe have to join it with inserted DNA so first we replace it again we put it to join with the DNA that we put there... hope its clear.

I think you switched between first isolated and first discovered...because it’s clearly written in ncert that Roman numbers are based on order of isolation

Your lectures are very clear , easily understandable and without scribbling anywhere on the white board. After the end of the lecture if one sees the white board it your lecture when read should be followed. Sorry to point out one flaw when you draw a straight line below the Binomial nomenclature of any plant or animal or prokaryotic bacteria.Separate line should have been drawn below the generic name and a separate line for a specific epithet. I really enjoyed listening to your lectures.
