You don't need an injection port. put two layers of foil on the top of your jar. Lift up the first layer and inject culture or spore into the jar. It's been working fine for me for years.

@@trump45and2zig-zags make a still air box. Its hard to explain, you get a tote and make two holes for your hand and get some pvc pipe or something your hand can fit through. Glue some of those big rubber cleaning glove to the inside and seal everything up. I put glade wrap over the top of mine so I can see better. Put everything you need in, and spray some rubbing alcohol in, then put the wrap over it and let it sit a while.

Pro tip. Use a one of those triangle stone bits and drill through like you would a step bit. Its just sand through so there is no risk of craking or breaking at all

I don't understand this comment. With no special effort, I've never cracked a plastic mason jar by drilling. Frankly, if I was doing a shotload (ahem) of these, I'd probably pop for a drill press and do 10 at a time.

10:45 it's also the perfect time to 'noc up a few more jars with that LC so you can save some time on turnaround. Instead of having to go from the tissue biopsy stage next time. Lovely video BTW! Makes me want to get growing again. So much faster than cannabis with zero electricity required and basically one brief moment of light just to start pinning? You can't beat that. I'm just super weary about trying to go myco-psycho again because the last go around I got arrested for unrelated reasons and wasn't able to return home for a few weeks. By that time I had 8 Rubbermaid tubs that were just an orgiastic death feast of aspergillus and black mold plus some contams I'd never even seen before. Ever since then I can't even keep brand new bread in this freaking apartment for 2 days without it molding. I'm terrified if I put new tubs in they'd have the same exact fate.

By doing the tissue biopsy you select the best genetics from the mycelium. A pity to hear what happend. Anyways if you esterilize properly Im sure your not going to have any problem.

I bought the HaBeuniver 20mm injection ports, the shorter grey ones that seem to be very common, and they'll fall right out of a 1/2" hole but a 7/16" is a perfect fit. Thought this might save someone buying the wrong bit. A brad point bit works great on the standard metal lids, btw. Great video, Ashley, as always.

Hey Ashley, check this out, this is from a fellow named Marc R. Keith who has been growing since the 70’s, “Throwing spores into honey or karo has to be the worst possible way to make a liquid culture and I'll be glad when this current fad passes and folks can get back to established techniques. Just inoculate some grains or brf in the standard way and you'll be miles ahead. For those who wish to make LC for inoculation, here's a way to get 100 syringes full of inoculant in two weeks: 1) Inoculate a quart jar of rye berries or wbs with your spores. Upon full colonization, shake the jar to loosen each individual kernel. Be sure to give it the smell test to make sure it smells like fresh mushrooms. 2) PC a quart of distilled water in a jar with a filter disk or tyvek, etc. In a glovebox or in front of a flowhood, pour 2/3 of the quart jar of sterilized water into your jar of shaken grains. 3) Shake well and then pour the now mycelium rich water back into whatever was left of your jar of sterilized water. Use the jar lid to hold back the grains themselves from pouring out. You now have a full quart of highly concentrated mycelium water that can then be used as is, or diluted two or three times again, and used to fill syringes. Because the grains were shaken first, the mycelium is ripped into shreds and will be sucked through even small needles easily, although larger needles always work better for this. Remember, mycelium grows poorly in water unless under constant stir which oxygenates it. With grains, they colonize in two weeks or less because oxygen is throughout the jar in the spaces between the kernels. LIQUID CULTURE - A glovebox is not advanced equipment. One can be made free with stuff around the house, so there's no need for people to work in open air or fecal and mold infested bathrooms and kitchens. There is also no way to generate 100 full syringes of liquid mycelium in very small pieces within two weeks from injecting spores into honey or karo. I'm sure if people blindly used the grain method I described above, many would still have problems too, because they're inserting unproved and possibly contaminated spore solution into grains, which also isn't proper mycological technique. However, agar can easily be poured in a glovebox, and the Petri dishes used to isolate healthy mushroom mycelium away from any contaminants in less time than it takes to even know if a karo LC is contaminated or not. That agar wedge can then be used to inoculate the quart(or pint) jar of grains, that can then be turned into at least 100 syringes, or used for g2g transfer to get ten jars the old standard way. In addition, after the water is poured out of the grain jar, it can be placed back on the shelf to re-colonize, and the process can be repeated again. I just strongly feel new growers are doing themselves a disservice when they simply inject spores into honey water and then sit there waiting for something to happen. At the very least, inject directly to grains so you can visually inspect the process. Perhaps Agar's lids with injection ports will help those who have never used grains to get off to a good start.” From, “All of RR’s notes.”

It's very proper but without the equipment it can work as well. I can use 5% agave syrup into sterilised water in a jar with a hole filled with silicon. Using small sterilised atones or glas marble in the jar to shake it 2 more times a week. Anyway, it's probably not as good as with this equipment but I can tell it works as well. If you grow a lot you probably improve your equipment anyway to minimise your failures and experience.

Amazon has those lids in colors, with silicone gaskets. They are sweet. I use 1/4 drill grey injection ports, and 1/2" stick on air exchange glued down with red high temp RTV sealer. They work great!

I’d like to know about the benefit of doing it this way versus just getting a sports orange because it seems like a lot of expense and cost to do it this way and as far as how long it takes, I don’t think you’re saving any time in the long run. Igrow mushrooms I've been doing it for about 10 years I would say I have no problem with infections ever on Agar. But it's a lot easier to get infections in liquid media for some reason I don't know why it just seems to be a lot easier I mean I have had probably 5050 infection to success ratio for liquid cultures that which is terrible.

The plastic lids do *_not_* seal against the jar. This may not be an issue for this process, but it's generally good for anyone using mason jars to be aware of this fact. Also, for people without a magnetic stirrer: An Oster blender (a.k.a. "Osterizer") blade assembly and sealing ring will fit a wide-mouth mason jar! Cheers from elsewhere on occupied Anishinaabe land.

Liquid culture is not spores it is live mycelium Yes a syringe of culture will work but spores need to germinate into mycelium so it has to go to agar/mycelium/ then to a LC She said this a few comments prior 👍

Instead of inoculating your liquid culture from an actual fresh mushroom picked from harvest, can’t you also inoculate using mycelium that you grew on an agar petri dish? Would all the steps be the same except for the what your syringing (viz your Petri dishes’s mycelium as opposed to a freshly picked split base of a mushroom)? Thank you and great channel!

I store them in my office. It is about 70-75F in there most of the time.

I want to echo everyone's thanks. These videos are excellent. Regarding breaking and shaking the bag, can you do this too often and if yes how many times is enough, 1,2,3...? Thanks

2 is more than enough, anything over that and you might risk killing the mycelium or putting it in stasis. I usually only need to shake once at around 35-50%

Are they placed in 33-36 f or fridge temp? Are they stored at room temp alongside monotubs? Does putting it in the fridge make it go dormant like it would a grain cake?

Any extra light what extract? I live in Sweden, I don't have access to the ready mixes so need to know how I can mix the medium myself. Sweden is still in the dark ages in much respect...

Hey thanks for the video! I have a question though. Say I buy a oyster mushroom liquid culture online. Can I inject that into my own liquid culture mix and have it duplicate? Thank you!
