Hello, thank you for the video. I would like to ask I have seen and read about using methylene blue or violet for the yeast cell count dead vs heathy yeast. is this possible ? and is there any downside doing this way thanks

Methylene blue works in much the same fashion as trypan blue, and the staining is performed in the same way. The only difference is that methylene blue can stain dividing cells as well as truely dead ones, and as such, more care needs to be taken in counting and in interpreting your counts.

So there was a total of 240 cells you counted? So you would have to count all 4 squares to get that number of cells?

Nope, I'm 100% digital. I show how to do counts like this in episode 10, which comes out Monday. There is also a good free app for your phone called hemocytometer sidekick

What magnification are you counting with....400x?

For yeast, 100x, which let's you see a whole grid area in one field of view. For mixed cultures I use 400x and the central grid.
