Thank you for the video. It is really clear and helpful. But I think there is a small problem concerning the ratio of blue: red fluorephore. First, if the blue:red = 0.75, then there is more red fluorephore, the color should be more red. But you used the blue pen to draw it.But maybe you didn't mean to correspond the pen color to the real color. Second, the second ratio of blue: red you wrote is 1:0. But I think it should be 1:1. Anyway, the video is very constructive.

Thank you for the easy explanation of that method! :) I have only two questions: Should the value be not higher than 1,25 in case of duplication? And the same thing, should the value be not lower than 0,75 in deletion?

Dear All There are two serious mistakes in this video! 1. In CGH experiments, we isolate the reference DNA from a healthy individual. But this guy argues that both the tumour and reference DNA are isolated from the same individual. This is a scientific flaw. 2. The tumour and reference DNAs are not hybridized to each other! We hybridize them on normal metaphase spreads! 3. The term that he uses "Metaphasic" does not exist. "Metaphase cells" is used instead. Please be aware that this video contains serious scientific flaw! But it is not surprising because everyone can upload videos on youtube.
@zedor15539 жыл бұрын
I believe that for clinical research, it is normal to extract DNA from tumor and compare this DNA with surrounding healthy tissue from the same patient. DNA from healthy tissue serves as control for studying new chromosomal abnormalities in tumor cells. I can't encounter the scientific flaw at this point, and I'd be really grateful if you could explain this point further. And, I would appreciate if you write your references if you claim that "everyone" can upload videos here. And, as well, anyone can comment on them. And "Metaphafic" is indeed an inaccurate term.
@ermanistifli80749 жыл бұрын
Dear Zhannie Could you please show me a study that utilizes the same individual as a source for both reference and test DNA?

more advanced information would be useful. how do the data look like? what is resolution? how are data processed (examples of data files)? also how many fluoriscent probes are on each chromosome? how do mutated chromosomes hybridize each other? can they wrongly hybrydize? how many chromosomes are there from each cells? 1 or are they amplified?
@pramitbanerjee8 жыл бұрын
the data is image files. You use certain software to quantify the fluorescence values, then you can use matlab to analyze them

I have a doubt...what about when we're talking of genomic diseases like for example cris du chat syndrome. From where do you take the 'normal' DNA? because all cells have this mutation. Thank you
@tomasgybel196410 жыл бұрын
I guess it is not appropriate diagnostic technique then :/ Maybe something like M-Banding, SKY or M-FISH is better for like "cursory diagnosis".
@r3g4rds10 жыл бұрын
you can use an array. So you can use a-CGH.

hybridization is not between the test and reference genomes (DNA), but between the microarray oligonucleotides and the both test and reference genomes (DNA).
