Culture media, types of media based on consistency/solid, liquid and semisolid media - Part I /Tamil

In this video, the general information about the culture media and its types are discussed.
A culture medium is a solid or liquid preparation used to grow, transport, and store microorganisms. To be effective, the medium must contain all the nutrients the microorganism requires for growth. All microorganisms need sources of energy, carbon, nitrogen, phosphorus, sulfur, and various minerals. Culture media can be classified based on several parameters such as Chemical constituents from which they are made, Physical nature and Function. Based on the Physical state (Consistency), culture media is classified as Liquid media, Semisolid media and Solid media.
Liquid media provide greater sensitivity for the isolation of small numbers of microorganisms. Examples of liquid media include nutrient broth, sugar media, and enrichment media. Broth medium serves various purposes such as propagation of a large number of organisms, fermentation studies, and various other tests. e.g. sugar fermentation tests, MR-VP broth.
The solid medium contains agar at a concentration of 1.5-2.0%
Solid media, provide isolated colonies that can be quantified and identified. Solid medium is useful for isolating bacteria or for determining the colony characteristics of the isolate. Nutrient agar prepared by adding 2% agar to nutrient broth is the simplest and most common medium used routinely in diagnostic laboratories. Other examples of solid media include blood agar, chocolate agar, MacConkey agar, etc.
Semisolid medium is prepared with agar at concentrations of 0.5% or less. Semisolid medium has a soft custard-like consistency and is useful for the cultivation of microaerophilic bacteria or for the determination of bacterial motility.
