The girl with the heavy make up is one of the Harajuku Gal style called Ganguro(Which means Heavy dark) which was a fashion statement against the Japanese beauty standard, and was very popular back in the 90s. They took hours to do makeup and built their own language inspired by English. From what I know, because they didn't want to wash the makeup away that they'd skipped taking showers or wash their face for days, and had bad reputation(at least in the media when I was growing up).

No, she's just being very crass, she's a Youkai that's sympathetic to rape victims so she defaults to cursing men to lose their junk. The tunnel she's in has had a lot of women raped and murdered in there, she's spending time consoling the victims whose souls are trapped there, so seeing a male come through, she went for traumatizing and cursing him.
