We use a standard ovoil, but we do customize our pipettes for better control.

In this video, we used a single laser pulse (400 micro seconds @ 100% power) that resulted in the cells separation from the trophectoderm. Without laser assistance we would not be able to remove the cells. The edge of the drop is used only to stretch and thin out the bridge between the trophectoderm and biopsy cells with allows us to minimize the number of laser pulses.

The hole can be 5-10 µm. Yes, freezing immediately after biopsy is acceptable.

In this video, assisted hatching was performed on day 4. Hatching can also be performed on the day of the biopsy on day 5/6.

Are you using this technique with every biopsy? Or do you apply the flicking technique as well? And regarding the technique show in the video, when you go to the edge of the drop, do you apply any suction from the biopsy pipette or do you just pull with the holding as I think you're doing in this particular biopsy? At RGI we try to use the edge of drop for most embryo biopsies. This approach allows us to make the cytoplasmic bridge between the sample and embryo as thin as possible. That, in turn, allows us to minimize the number of laser pulses to 1-2 or avoid them completely. To stretch the cytoplasmic bridge to its maximal we do both: pulling with holding pipette and pulling by the biopsy pipette simultaneously. We also use flicking, but in rare cases, when for some reason a laser cannot completely detach the biopsy sample from trophectoderm.

Per PGDIS guidelines, mosaic transfer recommendations are deferred to the provider. In our PGT lab, where we work with clinics from around the world, mosaics account for about 8% of PGT-A diagnoses.

Only about five cells are removed from the outer layer of the embryo. This layer will eventually become the placenta and therefore should not affect the development of the embryo. This technique is necessary for PGT.
