Seeing as Chuck built the Angels ans Archangels without any dna being involved I imagine them all like adopted siblings in human terms, so in terms of blood relation theres no connection. Which is why its ok when Cas and Hannah get involved with each other. Like Percy Jackson demigods, the dna doesnt exist so its all ok. So Cas is kinda Jacks dads adopted brother, but... i mean, the point of the show is your family is who you choose not just blood. Jack chose Cas as his father, and that makes Cas' husband his dad too
