To test the presence of albumin in urine
REQUIREMENTS
Test tubes, test tube stand, spirit lamp, test tube holder, urine sample, 30% sulphosalicylic acid,
PROCEDURE
1. Sulphosalicylic Acid Test: To 2 ml urine sample, add 2 ml of 30% sulphosalicylic acid and warm
White turbidity in the sample indicates presence of albumin.
*Aim:*
To detect the presence of protein in urine using the Sulphosalicylic Acid Test.
*Requirements:*
1. Urine sample
2. Sulphosalicylic acid solution
3. Test tubes
4. Dropper or pipette
5. Water bath or heat source (optional)
6. Centrifuge (optional)
7. White background surface
*Procedure:*
1. Collect urine sample: Obtain a fresh urine sample in a clean container.
2. Prepare sulphosalicylic acid solution: If not already prepared, obtain or prepare a solution of sulphosalicylic acid.
3. Perform the test: Fill a test tube with the urine sample to about half full.
4. Add sulphosalicylic acid: Using a dropper or pipette, add a few drops of sulphosalicylic acid solution to the urine in the test tube. Mix the contents gently.
5. Observe for turbidity: Observe the mixture for the formation of turbidity or cloudiness. If protein is present in the urine, it will cause the solution to become cloudy.
6. Centrifuge (optional): If desired, you can centrifuge the mixture to separate any precipitated protein. The supernatant (clear liquid) can be poured off, leaving the precipitate at the bottom.
7. Compare with control: As a control, you can also perform the test with distilled water. Distilled water should remain clear, without any cloudiness.
*Results:*
Positive result: The formation of turbidity or cloudiness in the urine sample after adding sulphosalicylic acid indicates the presence of protein.
Negative result: No cloudiness or turbidity forms, suggesting the absence of significant protein in the urine.
