Sir how we know about primer sequence, as we don't know about dna sequence???

We clone the dna strand which is to be sequenced. And the sequence of vector we're using for cloning is known. That's how primers are constructed.

We didnt add the terminator nucleotide in normal form thats why as the terminator comes with its complimentry the strand stop extending..and the next strand start from the same site where the fist termination occure.....
