YES YOU CAN.. YOU NEED TO MODEL TX AND RX ANTENNA, THEN CAN CALCULATE S21... You can refer V2V video in this channel

Use plane wave excitation in antenna and terminate antenna port with 50ohm. Then integrate E.dl across the 50ohm ( using field calculator - find field calculator cook book from internet), you will get voltage generated across the antenna port

You can run RF discharge analysis is HFSS or multiplication analysis, depends on where this antenna finally going to use (in atmospheric condition or space)
