A bunsen burner/gas blowtorch could also be substituted in place of the laminar flow hood if that is all you have at home. As someone who did some work in an old school bio lab we used to use bunsen burners to create an updraft of air to prevent airborne bacteria from landing on our sterile items. Obviously you need to be careful using this method but it is quite effective

Ive heard of this method before. I've also heard it's not a very effective method of environmental sterilization. Even the particulate matter from the Gas used could contaminate your work. A better option for at home DIY method is using a "still air box".

Funny thing though I hated science at high school growing up , I like the hands on stuff theory was to exhausting lol , in my older life I quite enjoy the science of things when you understand the why and what for lol , things at high school just seemed to take forever and I didn’t have the patience then but now I’m even trying mushrooms which is harder than you think with contamination etc , great video .

orchids grow in the wild without all that stuff, how do they do that?
@janky200
@janky200 10 ай бұрын
Via symbiotic relationship with microfungi which provides sugars for growth as well as protection against bacterial infections! Orchids seeds can be very sensitive & as some orchids evolved they developed a back up plan to reproduce via growing little babies at their bases or off of their flower stems, much easier for them then their little seeds all failing because conditions aren't right!

For anyone trying this at home. Watch some mushroom growing videos! And if you dont have access to laminar flowhoods you could always make a "still air box" using a clear plastic tote from any hardware or basic goods stores world wide. Make 2 holes into 1 side of the tote large enough to fit your arms inside. Clean the area very good and all over. Inside the tote you could also use a spray bottle to mist alcohol inside the the upside-down tote. This will sterilize the air inside the tote. (Make sure you have everything you need inside the tote before finish cleaning everything. Dont introduce unclean items into the tote and if you do you need to repeat the process over.

Just before adding the charcoal, how much liquid was in the beaker? Not all of that 1L stock solution was used, so I'm curious about the charcoal-medium ratio. Cheers

Can anyone explain to me, If orchids need such sterile conditions to germinate, how do they manage to do it in nature then, where conditions are opposite of sterile? I know they get their food from fungi in nature, but why don't they do the same thing in laboratories to avoid all the hussle with sterilization?
@shannona6989
@shannona6989 Жыл бұрын
They don't need sterile to germinate. There is thousands if not million seeds in a single pod, literally like talcum powder, they haven't proteins like a normal seed just an embryo, they scatter forever in the wind with the chance one lands in the right spot with the fungi to assist germination. In a flask you can germinate almost 100% of the seed without the need of the fungi, because they take so long and need such a specific media to grow in it has to be sterile as it's also the perfect environment to have bacteria bloom. Which grows super fast and smothers out the orchid seeds which can sometimes take months to germinate.

@Agriculture Academy, could you recommend a method of storing seed pods ? thanks
@shannona6989
@shannona6989 Жыл бұрын
Dry place, not in a place where it will sweat or rot. You've very limited time to use the pod, same as with dry seed. The longer it goes the more chance of bacterial infection where it gets to a point the seed becomes to dirty to clean and use.

Orchids have a symbiotic relationship with fungi in the wild - these fungi provide the seed with sugars so they can grow. At least that's the simplified explanation

Orchid seeds dont keep so theres not really a market for orchid seed (thoughtl there are a lot of scam sites). This kind of thing is really just for if you yourself want to hybridize or cross breed orchids that you already own.

Lol growing orchids from seed is not a casual endeavor, regardless of your wants. If you dont already know how challenging it is to grow orchids from seed, maybe you should just stick to already grown orchids for a while, or propagade via keikis.
