Do you need to disconnect the brake lines for the 3.25 springs. I’m currently installing the pintop kit and axle lower enough to get them in like this video.

No, just make sure the axle is supported so it isn't hanging from the driveshaft.

Are re location brackets necessary for the track bar

We usually prefer to just to an upgraded adjustable length bar. Drop brackets are common because they're cheap, but then you're stuck with the stock track bar which isn't ideal.
