If transmit antenna side is spun instead, the result pattern will be the same?

If both antenna is dipole and measuring omnidirectional, then yes. Normal pratice only receiver antenna is rotating.. But you also can only rotate transmitter antenna, and left the receiver antenna stay static.

how to calibrate the entire system for every power levels in your test setup?

For the power level calibration, just need to calibrate once, the step is adjust each power and save it in excel file, then load into the software. The radpat also have function to perform this calibration with step and automatic save into the software. You may refer to the video description to download the lab module sample from dream catcher, inside have shown the step of calibration.
