I'd remove chain pop it in to a zip bag, rear derailleur and mec 100%, deflate wheels and leave tubes open, place black electrical tape over the break blocks once inserted to ensure that they don't fall out, rotors take them off, loosen headset on fork, mark seat post and remove saddle, bottle cages off and have several zip seal bags for components and parts... then pack bubble wrap over the frame after wrapping a few microfiber cloths over the frame... these tasks take next to no time but will protect the frame and components spending an extra 30/60 min doing this is well worth the effort.

overkill, you can just use a chain mount and strap the bike in properly

looks like it's a 52, these boxes are great but damn don't follow the steps here some very important and precuationary details left out for sure... Long term storage isn’t recommended but short duration travel isn’t an issue.

Hmmm ok well travelling U.K. to Spain so will be upside down maybe 4/5 hours
