I use the cling wrap like you. It takes two seconds to cut little rolls with my band saw. Not everyone has a nice band saw though.

I just put a swab I received from much onto plates. I wish I would have watched this video 1st. I just rubbed the swab over the face of the arag. I'll be doing this method on my next swabs for sure. I'm assuming that when you shove the cotton into the agar at the end that it creates a 3 deminsional growing environment around it?

Yes. I think it a better way to use the whole matrix of agar. The mycelium will grow out of the cracks and spread over the surface. Then you can subculture it to a new plate.

as a noob wrapping plates with clingwrap sections in the SAB has been the hardest part. too poor to justify parafilm, that new grafting tape any decent? been wanting to try one, would love a brand recc

You can take the plates out of SAB to wrap them. Just make sure you hold the lid tight while wrapping and I wouldn't set them down anywhere that might be dusty. Do them one by one after you finish your transfers. My friends discovered a new brand of grafting tape that is way better. Same with cling film, just try different brands. Some are better than others and it's kind of personal preference. I like mine on the 'sticky' side.

why do u dig into the agar instead of swiping the surface?

The spores will germinate better if they are protected a bit from the 'dry' air. Mycelium will shoot out of the crack. If there is bacteria on the swabs, the mycelium will grow and hopefully leave it behind in the crack. Plus, it's just easier. The spores will definitely be ripped off as the cotton is pulled through the agar.

Very short answer. Monokaryons are haploid (n) gametes. Dikaryons are n+n. Google is probably better.
