I bought a surplus DNA Sequencer for under $100! I don't know how to use it yet, or if it's even fully working, but it's still pretty cool! In this video, we do some unboxing and initial tests to see what it has and what it might need.
The system is an IntegenX Apollo 100 from around 2009. It includes the lab-on-a-chip sequencing units, fluid-handling Tecan robot assembly, pressure and vacuum module, control computer with software, and some other bits and pieces.
While I don't yet know how to clone any extinct animals, create mutant monsters, or even check my ancestry or geneology, I can still say I own a DNA sequencing lab! Stay tuned for future videos when we (hopefully) figure out more about what this thing can and can't do, how to operate it, and what we might need to do something interesting with it.

This is awesome!! I'm a bioengineering PhD students so I'd have a blast playing with something like this! I do genetic engineering with yeast. From some reading, this machine does Sanger style sequencing of around 900 bases of DNA at a time, which is what my lab still uses. We pay about $3-4 per sample to have them sequenced with next day results. But I've always thought in-house sequencing would be way better! In order to do Sanger sequencing, you need a concentrated DNA sample (you likely couldn't use your whole genome) and a primer, which is a short (20-30 base) fragment of DNA that tells the reaction where to start reading from. You probably need to make a few different buffers to run the machine as well. I'd be happy to help you get it set up and send you some samples to sequence! We couldn't sequence your whole genome, but we could sequence a fragment of one of your genes pretty easily. I think it'd be a fun experiment!

Interesting! I've talked to a couple people who are somewhat familiar with this unit or things like it, and so far it sounds like it would be complex and expensive to actually do much with it. It didn't come with any chemicals or primers or anything.

Hi, I have a Beckman Coulter 608450 CEQ 8000 Genetic Analyzer System, I was hoping to use it to do dna methylation test, would that be feasible? would it be capale of doing geneologiccal studies?

Just incredible. I finished my degree in Biochemistry in 1991. This would have been worth millions in those days. It would be another 8 years before human Stem Cells were accomplished. My husband was treated with them in 2020 for multiple myeloma. In another 30 years I guess we will be able to treat anything for the rich anyway.

I did a follow-up on this where I determined it's kind of useless without expensive chemicals and supplies. Sadly I can't bring back the dinosaurs, I even asked a few scientists and they thought it was too old to get going on my $0 budget. I ended up trading it to a viewer for some other stuff.

Old DNA analysers are useless, the tech has moved so fast that even if you could still get the consumables they would be way more expensive than modern services. And even if you could use it, you'd still need a lot more lab equipment and trained operators to do anything useful. I've taken apart several - one originally cost $500K, and consumables were about $20K per run. Think I paid about $300 for it. it is interesting to see how they build gear that is basically "cost no object"

7:12 Same with me, they forgot (although I feel dumb for not noticing, yet checking for EVERYTHING ELSE lol, gotta love adhd sometimes) not only the 3 or so serial port cables, BUT ALSO THE RUDDER PEDALS. I’ll either hack together something from an Arduino (some good diy/os designs exist already), or pay 100 or so dollars to get the “real deal” as an oem replacement part. Either way CHECK FOR THINGS AND ASK WHEN DOING THESE. Otherwise, as stated in my other comment, i 100% recommend going to these.

Yeah that would be the last resort, if I can't use it I'd try to give it away. But sometimes old tech gets so old that no-one even wants it donated, like the wheelchair lift that was too sketchy (so I made it into a tank).

@@saveitforparts Also look up if you have any local Makerspaces/biohackers, worse case scenario send it to someone like “TheThoughtEmporium” etc. Kit like this is an AMAZING find, and will be a huge help to those who know how to use it and need it!

@@saveitforparts I was about to say; before ripping it apart and dooming it to junk giving it to someone in your area who knows how to use it is peak chad move. Especially given how much the American tax payer already paid for this. It's a disgrace it ended up in your hands for $85. That is an indictment against the overspending and waste of big government.

Yeah, University Surplus Stores are awesome! COVID threw stuff into a more hassle ways and means... and seems pricing went up other than the auction stuff at times. Looks like that blue thing holds that larger Nalgene bottle? Maybe was never used? I had equipment that before I implemented the Design Qualification (DQ) into the Equipment Lifecyle SOP... the lab would purchase equipment they never used since didn't meet the lab specifications test procedure requirements and then would just list on the lab auction site eventually (I forget which one Perrigo and I think Pfizer used since I didn't deal with those systems). Will be interesting to see what you can determine to get working cost effectively. Would be interesting if you can find the software and/or hardware engineer(s) and/or scientist(s) that developed the equipment? Maybe even a lab tech might know enough?

Tear out the XYZ controller and install arduinos with motor drivers for a small CNC or 3d printer. The mechanical part of that system has got to be better than most hobbyist units on the market. Tecan is a well known biomedical engineering company that collaborates with numerous others in the field.

Try reaching out to The Thought Emporium. See if he's interested in it or could at least point you in some direction on what to do with it. He's in Canada so shipping costs would be insane but I know he has lots of connections to groups that do genetics. Some in the US.

I am looking forward to the dinosaur park addition at sandland. Also since I took my A+ certification back in 2000 yes I cannot forget serial ports, or SCSI device termination and ID mapping. Speaking of weird stuff and professional hoarding... you happen to be interested in a 24' diameter 52" depth Intex pool liner? Used for a couple weeks then stuffed into my shed after it tore along the top rail. Have pics of the damage and part number if interested. I am in west st paul.

Just can't bring myself to chuck it because it still holds water but is scary as an above ground pool due to the support being compromised. Maybe I will turn it into an in-ground pond? Or maybe I should hold on to it until you get more mileage on the DNA sequencer because it'd probably make a great water dish for the dinosaurs. 🤣

If you have the yard space you could do a garden pond, we made one with a stock tank and some extra plastic pond liner. It brings in a TON of wildlife, we see all sorts of cool migratory songbirds and other critters now that we have a pond. If it's 52" deep you could dig it below the frost line and over-winter fish in it.

GPS output for navigation software was serial bus for years! Post- LORAN. Is the sequencer essentially an auto PCR/electrophoresis?

I was recently involved in provisioning a bunch of $4,000 laptops that had built in GPS. Brand new models. They still use serial to communicate. Granted, it's all internal but I had to make sure they were assigning the correct COM ports to talk to software.

I'm still not 100% sure what the thing does, apparently it handles the first two steps of a Sanger sequence (but not the electrophoresis part). It might be a little less useful than I thought, but I'm consulting with some more knowledgeable folks to see if we can do something with it!

Interesting! Only time I really use serial these days is when I have to program some oddball device like a radio or one of those RV satellite dishes. I have a couple other things with mysterious serial ports but no documentation on how to talk to them.

Dumb and maybe 1 good ideas: Homemade 3d printer. Personal Floppitron. 1st request would be "Home Improvement" theme. Empty one of the cabinets of parts, put a shank in it, fill with concrete. Literal boat anchor for the jetboat. Clone a condor or ibex instead. That way you won't have to hear, um, Jeff Goldblum go on about chaos.

@@saveitforparts Yeah I'm from the UK and have lived in Spain and Poland, and .. I dunno, it's just not "a thing" here for all sorts of stuff like this.. At best, we get gov't stolen cars and bike auctions.. the rest, as far as I know, gets insolved privately at the list price or just gets junked..... Plus no doube we simply aren't even allowed to deal with ex-lab stuff like that publically.. Also, we just don't have the space for it all.. Sadly, most of our stuff gets compacted and landfilled - you said something in this video I've been saying for a long time - DO NOT THROW STUFF AWAY.. however, first you gotta get your own bit of land and space, and then stand and protect that space til you die.. :/ Over here, everyone's getting shoveled into holes and boxes real quick.. :(

Yeah, people can ask whatever they want on eBay, you have to look at the sold items to see what things are actually worth. I just saw another local auction with one of these and some other lab stuff. I didn't bother bidding and it went for like $6 :-P

What kind of cat is that you picked up? Our cat looks exactly like it. We found ours when it was about 4-5 weeks old in our yard nobody claimed him. Definitely looks like a pure breed

Both of our cats are 2nd-hand, unknown breeds. Donny (the shorthair) kept showing up in my garage, we found the owner but after about the 5th time they said he must like me better. Fluff (the fluffy one) was from a household with too much drama between other cats. They get along OK with eachother most of the time!

@@saveitforparts Yeah that's a good point, still though I would bet its worth more than just $100. I hope I can find a good deal on one once I get enough space to setup a proper bio lab. I really want to be able to sequence my own dna and test it for a certain gene that runs in my family which causes peripheral nephropathy, I am hoping by the time I have kids I can find a way to remove it if I am a carrier.

Sadly i could not fit the *Forensic Cyanoacrylic Glue Fuming Chamber* in my 90s honda accord (but did fit the flight simulator console…WITHOUT REMOVING ANY SEATS LMAO, sketchiest drive home though lol), otherwise i would have had the world’s BEST 3D Printed part smoothing machine… Either way i 100% reccomend these if you have some money to burn, ESPECIALLY if you share the fun at Makerspaces etc (as i hope to do one day) not hoarding it yourself, or worse end up selling them for more…

Vista good. Before they meddled with the usb stack and made windows non medically compliant. (Latency ..remember how laser mice saved us from terrible motherboards...wasnt the boards, was Microsoft) (Details in the microsoft white sheets for the usb stack update)

To sequence DNA on these old machines it will cost thousands of dollars. That's why these are absolute. New machine are way cheeper to run!

Actually, if you’re willing to 3-D print the custom plastic parts that are single use and can purchase the chemicals used individually it is way cheaper

for a dna sequencer especially back then you're looking for like half a million dollars plus its still probably worth 10 g also sell the computer with the machine the computer should have specialty software on it

It depends on the area, there are some like publicsurplus that cover most of the US and others that just do one state, and then tons of little websites that just do specific cities. There are some for estate sales, some for surplus vehicles, some for business liquidation, it really depends where you are and what you're looking for.

I traded it to someone for some old solar panels and an old 3D printer. Couldn't find the chemicals it needs, and it was too old to really do useful science.

Windows Vista. Those were the days. Junk OS. Pure crap. Nice try with the revival. Dang thing sounds like a printer from the mid nineties. By the way, that was a parallel port, not a serial port.

It's the size of a parallel port but uses serial protocol, not really sure why they did that. It works with a generic USB-to-serial adapter. I kind of dimly remember my old DOS and LAPLINK experiments with 286 laptops, and I don't think parallel and serial were directly compatible without additional converter boxes.

There is a great scene in The IT Crowd where Moss is standing next to a bomb disposal robot examining a suspicious package. He's really just quite excited by it all. ​​@@saveitforparts if I remember correctly, they both could use 25pin 'D' centronics connectors, but the gender would be reversed for serial, I can't remember which one was was male or female for peripherals, my old printers aren't handy, and I often mixed them up when I used them often.. SCSI one used that connector too! Got a scanner that needs a 25 to 50(or68) pin adapter..

I traded this one away, sorry! You could search for online surplus auctions in your area, that's how I found this originally. Or if you live near a college, they often have stuff like this for sale from a surplus or re-use program.

Some say peak capitalism is bad, but I think it's pretty cool that ordinary people can get newish in box high end biotech gear for under 100 bucks. Society may collapse but us tinkerers, makers, and mad scientists will be entertained learning things 😂.

📺💬 DNA test sequence 🥺💬 It is sequential differentiation you know in AI but it is a long string and does not exactly match they need to compare in segments but I watch from the VDO the tools are new for me. 🧸💬 Not only DNA but it can test some compound's properties if they are capable but the main objective is how you map large long strings with small tiny pieces like compound DNA. 🐐💬 I think identity is not enough if you think a DNA test is a must. 🐑💬 As the same other tools otherwise you becomes a doctor nephews.
