Some targets are difficult-to-detect mainly due to the facts: 1) improper antigen retrieval may cause false-positive or -negative results; 2) some antigens have lower abundances; 3) some available primary antibodies are less efficient/specific; 4) many other parameters in the procedure may also significantly affect the detections; 5) successful quality IHC detection requires a proper combination of optimized conditions; 6) data analysis and interpretation require integrated knowledge and skills.
