DNA polymerase require a primer to start DNA Replication. Without a primer template along wit with a free 3'-OH, DNA polymerase can't synthesise de novo. DNA polymerase catalyses a nucleophilic attack of by 3' OH on the alpha phosphate of the incoming complementary nucleotide. Thus it needs a primer to begin with. Whereas, RNA polymerase can make RNA without any primer. So initial primer is made by RNA polymerase which is later removed by DNA polymerase I by its exonuclease activity which allows it to do nick translation. As the DNA ligase doesn't seal the nick between the RNA primer and the DNA, that nick can be used by DNA polymerase to replace the RNA primer. But for PCR or any other in vitro assay, where we order primers, we use DNA. As Taq Pol or any other PCR polymerase can efficiently polymerise irrespective of DNA or RNA primer.
