This is an example of the right circumstances to take the risk of the side effects of any immunotherapy or gene editing or therapy. Also the slow and safe speed of such gene editing. If you look at the side effects of these things. They are acutely familiar to other technology and also any Interruption in the immune process on that level. Not that this isn't amazing work. It truly is... This presentation is presented in a way that skips much of the luck against casino house odds that go with all of this in any certain individual. The age of this girl obviously has much to do with the success so far as children are much more resilient and adaptive. Which I imagine is the reason so many experimental approaches on children are free despite how it's afvertised in a more pliable.. stem of study. Yet youth a very fine line both ways that can fall off the tightrope being walked here. Defending on which responses, profilarion after responses are effected by many factors. Adaptive modulation signaling problems are varied and not fully known. Especially on the nuclear level in this system... but both combined.. of millions of variables of individualized possible reactive responses and counter responses. Hence the not understood neurological problems. One could guess.. If you compare this to the way the mRNA delivery was so just completely recklessly and quite frankly completely carelessly when you compare all the time, care and caution used in simular approaches in immunotherapies for cancer. The risk in approach in such profilarion are to me an incalculable order of magnitude higher by any honest comparison of really the only comparable tech that has any amount of the close appropriate years of study. Yet is still very very novel in comparison to all other timelines of any real effective treatment that has had success of longevity in the long term risk reward.. long term side effects escaping .. in the history of cancers medicine and vaccines. The role in the immune systems fight against cancer is somehow always left out of the conversation. Yet the market has been pushing it forward at light speed. The warp speed of profit. Instead of the careful methodical attempts like these that are expensive because they are being so cautious because of the tight rope of risk it involves and the after care and monitoring it takes during the whole process. The incrementally cautious and studied proper way of this sort of science.. Anything else is.. uncivilized.. I have been saying this since the very first mention of the mRNA tech being pitched for something that is comparable to a really bad flu virus. Because of my studies into the immunotherapies for my Dad's terminal cancer that could not get approved as recently as 2018.. 19.. Science didn't advance that fast to change the risk factors.. I was highly censored. Because if you compare this as any real medical science should or would not only would... You would find answers. Many answers that point out very obvious truths and answers that are being suppressed for the very reason they already know what these comparisons point out. Immeasurable recklessness based on all previous history of this science. In comparison to how the proper approaches like this can be. In the proper amount of time. Which gives me hope for this particular girl. Nice overview of a very complex treatment. A couple of hopefully useful suggestions and points. It’s often difficult to use ‘simple terms’ to describe the effect of a treatment. In this case perhaps disease free remission is more accurate and appropriate than cured. CD7 is expressed on NK cells and virtually all stages of T-cell development. This means as long as the modified CAR T-cells persist the patient will likely have impaired immunity. I really hope she remains in remission and has a happy long life. The diagram you give with CD28 stimulation coming directly from the cancer cell, is a bit misleading. Conventional T cells are activated for proliferation, ‘primed’, via T-cell CD28 interacting with APCs, expressing surface B7 family molecules, in secondary or tertiary lymphoid tissues. You correctly stated that CAR T-cell receptor DNA can be modified to have ‘intrinsic CD28 signalling’. This obviates the need for ‘conventional’ CD28 co-stimulation (signal 2) priming, in lymphoid tissues. Your diagram could be improved by replacing the cancer cell with an APC and then adding a CAR T-cell receptor diagram, showing the additional CD28 signal region and explaining how this replaces conventional priming. A small, but important point, CAR T-cell receptor DNA does not, usually, integrated into the genome.

Since the modified T-cells don't have CD7, what happens if there was an error in the coding creation of these cells or the base pairs that ends up making things worse? Is there a fail-safe in their design so that the modified cells can be instantly and totally removed or killed? If there is no fail-safe in their design, then it could become a horrific situation... A modified immortal cell line could be injected that could make a person a slave, like the Jem-Hadar in Star Trek, forever dependent on a drug to live, that is only available to those that follow their master's orders.

BUT I result to criticism for your failure to officially declare and document that which is clearly embedded in your conscious recognition of the DATA. This is my written that you are clearly aware of coronavirus related data, though you refuse to "speak your mind" and appropriately inform the millions of people, from whom have developed a preference to use your source for an important information resource. I can only assume that you have perused data acquired from KZbin, interpreted from twitter and other such "social media" vectors and comments, and long ago made a decision. You decided to ride a fence. Following your more recent multitude of informative presentations, I cannot assume such a decision was for altruistic reasons. I remain...disappointed.

I’m a Car-T cell patient and am in strong durable remission from mantle cell lymphoma! Thanks be to God. MD Anderson is where I went and it was wonderful. MCL is a type of large B Cell lymphoma-a Non-Hodgkin Lymphoma. 🌡️🧬✝️
