He could sense his Ki but was expecting an invisible fighter, so when he struck it caught him off guard because he hit exactly where he was but missed and then was taken out. Even 17 pointed out that he doesn’t have wings or fly so it’s not like Piccolo could hear any distinct bug/flying sounds he just knew someone was there. When they put it together that it’s tiny and jumped before attacking they used that against him and won.

Listening wouldn’t have helped Piccolo much in his situation because he was expecting a regular sized fighter and would’ve hit him with that strike had that been the case. I’m sure he would’ve figured it out if he had more time to react and figure it out, like 17 did since he wasn’t in combat.
