A lot of times enemies leave the room for the secret chest, especially slimes. And then when exploring later, you kill a random slime and all of a sudden it tells you that a chest appeared. You just need to figure out which room it was from lol Boss is always at the bottom of the layer, the bottom is one giant line of rooms with Boss door in the center one.

Wanted to give this game a shot but it always crashed between character creation and game start, had to refund unfortunately.
