I think the smile was adorable. Cas didn’t understand what he did to make Dean laugh so much. His smile was like empathetic smiling. At least that’s how I interpreted it.

I think he just has a strong sense of sarcasm and it's his way of being playful to poke fun. But I agree with the other commenter, that Jensen seems more intimidating to meet. I think that is because he isn't so easily able to get into his goofy place as it is for Misha and Jared.
