If you look back on the scene in Supernatural where Dean and Cas run out of the strip club and Dean is laughing but Cas is trying to be serious you'll see that Misha was smiling. He tried to hide it but I saw it. xD

I think the smile was adorable. Cas didn’t understand what he did to make Dean laugh so much. His smile was like empathetic smiling. At least that’s how I interpreted it.

I think he just has a strong sense of sarcasm and it's his way of being playful to poke fun. But I agree with the other commenter, that Jensen seems more intimidating to meet. I think that is because he isn't so easily able to get into his goofy place as it is for Misha and Jared.
