Not really, it has no effect on the vocal quality. However, if you just wanna feel safe. You can click on the channels and take them back to musical mode once you set the tempo.

You are welcome, you can right click on the project file then go to open with and select Cubase 10. Alternatively you can open Cubase 10 click on File top left corner then go to open and find your Cubase 5 project and double click it. However, you need to know that some plugins from Cubase 5 are not support in Cubase 10 so if you used any of those plugins in your project chances are that Cubase 10 might fail to open that project until you remove those plugins. For example the plugin Q
