All of it is cleary a joke between two best buddies and since their wives are besties too no one will be mad about it x'D

Fans always defend Jensen from "big bad destiel shippers who ask rude questions" but implying your screwing your best friend's wife is an enormously disrespectful joke, both to Gen and Jared. You can see that Jared is actually pissed for a few seconds. He has that, "I'd punch you in the face if there weren't a hundred people recording this." Yes, they are friends, but even friends cross the line. Jensen crossed it here.
